General description

The BioTracker Far Red Mitochondrial Superoxide Live Cell Probe detects superoxide, a reactive oxygen species (ROS) produced in mitochondria by electrons leaking during redox reactions. Cellular control of ROS is important because they are associated with disease, cellular damage and senescence. SCT278 is oxidized by mitochondrial superoxide and accumulated in mitochondria.

Add 10 μL of DMSO to each tube containing SCT278 (100 nmol) to create a 10 mM stock solution. Dilute stock solution with culture medium to create a 10 uM working solution.

Mitochondria are important organelles that uses oxygen to synthesize ATP, producing the necessary energy for living cells to thrive. Increased mitochondrial activity and mitochondrial dysfunction are associated with cancer, aging, and neurodegenerative diseases such as Alzheimer′s and Parkinson′s diseases. Mitochondrial mass (MM), mitochondrial membrane potential (MMP), and mitochondrial ROS (mtROS) have been widely studied as promising targets for mitochondria-related diseases. Since these mitochondrial attributes are dynamic, simultaneous analysis using multiple staining in a single sample confers a significant advantage.

Other red fluorescent reagents have widely been used to detect mitochondrial superoxide. However, their emission wavelength is a shorter red, which overlaps with other membrane potential detection probes such as TMRE, and is therefore not applicable for simultaneous staining with other mitochondrial markers in a multiplexed sample.
