Thermoacidurans Agar

Thermoacidurans Agar is a selective growth medium. It is recommended for culturing and isolating Bacillus coagulans (Bacillus thermoacidurans), a facultative thermophile. This bacterium can thrive in temperatures ranging from 20 to 55°C and at pH levels between 5.0 and 7.0. Thermoacidurans Agar contains proteose peptone and yeast extract, which provide essential nitrogenous compounds, vitamin B complex, and other vital nutrients necessary for bacterial growth. Additionally, dipotassium phosphate is incorporated to buffer the medium, ensuring stable pH levels during incubation. Dextrose is a primary energy source that promotes the development of B. coagulans.

Thermoacidurans Agar is specifically utilized for the isolation, cultivation, and enumeration of Bacillus coagulans (Bacillus thermoacidurans) from canned food products, as they are known to cause flat-sour spoilage in low-acid foods packed in hermetically sealed containers. B. coagulans will form large, cream to white colonies.

Suspend 39.2 g in 1 litre distilled water. Heat to boiling to dissolve the medium completely. Sterilize by autoclaving at 121°C for 15 minutes. Cool to 50°C and pour into sterile petri plates.

The designations basic, plus, or prime are added to indicate the quality control level, from basic quality control to standard QC plus to prime for full regulatory compliance.
