General description

MacConkey-Agar (without salt) is a powdered, non-sterile differential medium designed for microbiological applications. It is formulated without salt and crystal violet, making it suitable for culturing a wide range of microorganisms. The medium consists of agar, bile salts, lactose, neutral red, and peptone, which provide essential nutrients and selective properties for bacterial growth.

Application

MacConkey-Agar (without salt) is used for the detection, isolation, and differentiation of coliforms and intestinal pathogens, including Escherichia coli, Salmonella, Shigella, Proteus, and other Enterobacteriaceae. It is particularly useful for culturing urine specimens that may contain large numbers of Proteus species, as the medium restricts their swarming behavior. This media is ideal for clinical testing, environmental monitoring, food and beverage safety, and general microbiological research.

Preparation Note

Suspend 47 g in 1 litre of distilled water. Bring to the boil to dissolve completely. Sterilize by autoclaving at 121°C for 15 minutes. Pour into sterile petri plates. Dry the surface of the gel before inoculation.

The designations basic, plus, or prime are added to indicate the quality control level, from basic quality control to standard QC plus to prime for full regulatory compliance.
