General description

MRS Agar, also known as DeMan-Rogosa-Sharpe Agar, is a microbiological medium specifically designed for the enrichment, cultivation, and isolation of Lactobacillus species from various materials. It is a non-selective medium with a low pH that favors the growth of Lactobacillus spp. and Bifidobacterium spp. The medium contains essential nutrients such as glucose, meat extract, yeast extract, and peptones, which provide nitrogenous and carbonaceous compounds, amino acids, and vitamins necessary for microbial growth. Sodium acetate and ammonium citrate enhance selectivity by inhibiting the growth of unwanted organisms. The media contains polysorbate (Tween 80), acetate, magnesium, and manganese, which are known to act as specific growth factors for Lactobacilli, and provide a rich nutrient base.

Application

MRS Agar is widely used for the detection, isolation, and cultivation of Lactobacillus spp. and other lactic acid bacteria from food, beverages, and environmental samples. It is applied in agriculture, food and beverage quality control, environmental monitoring, and microbiology research. The medium supports the growth of fastidious Lactobacillus species, such as Lactobacillus brevis and Lactobacillus fermentum, and is often used in CO2-enriched atmospheres to enhance growth.

Preparation Note

Suspend 61.15 g in 1000 ml distilled water containing 1 ml polysorbate 80 (TWEEN® 80; Sigma P8074). Boil to dissolve the medium completely and mix thoroughly. Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes. If necessary adjust the pH with glacial acetic acid after sterilization.

The designations basic, plus, or prime are added to indicate the quality control level, from basic quality control to standard QC plus to prime for full regulatory compliance.
