TBX Agar

TBX Agar (Tryptone Bile X-glucuronide Agar) is a chromogenic medium designed for the selective detection and enumeration of Escherichia coli in foodstuffs, animal feed, and water. E coli cells absorb x-glucuronide and the intracellular glucuronidase splits the bond between the chromophore and the glucuronide. The released chromophore gives blue to blue-green coloration to the colonies. The peptone provides the essential growth nutrients to the organisms. Bile salts mixture inhibits Gram-positive organisms and the high incubation temperature of 44 °C further suppresses non-target organisms.

TBX Agar is widely used for the enumeration, detection, and identification of β-glucuronidase positive E. coli from food, feeding stuff, and environmental samples in food production and handling. It is particularly effective in differentiating E. coli from other coliforms, making it a valuable tool in microbiological testing for food safety and water monitoring.

Suspend 36.6 g in 1 litre distilled water. Heat in a boiling water bath or in flowing steam until the medium is completely dissolved. Autoclave at 121°C for 15 min. Cool to 45-50°C in a water bath, mix gently and pour 15 ml in sterile petri dishes. If stored at +2 to +8°C and protected from light the plates are stable for 1 week.
