General description

Mueller-Hinton Agar 2 is a selective medium recommended for the disk diffusion method of antibiotic susceptibility testing. It is formulated for testing rapidly growing aerobic or facultatively anaerobic bacterial pathogens. The medium is nutritionally supported by beef heart infusion and acid casein hydrolysate, which provide essential nitrogenous compounds, vitamins, carbon, sulfur, amino acids, and other growth nutrients. Starch is incorporated into the formulation as a protective colloid against toxic substances present in the medium. A key feature of Mueller Hinton Agar 2 is its controlled composition: it is manufactured to contain low levels of thymine and thymidine, which are known to inhibit sulfonamide and trimethoprim activity, and controlled levels of calcium and magnesium, which can interfere with the activity of aminoglycoside antibiotics. This precise formulation ensures reliable and reproducible antimicrobial susceptibility test results.

Preparation Note

Suspend 38 g in 1000 ml of purified water. Heat with frequent agitation and boil for one minute. Sterilize at 121°C for 15 minutes. Cool to 50°C. Mix gently and dispense into sterile Petri dishes.
