General description
Abberior CAGE 635 is a caged dye which is initially colorless and nonfluorescent. When illuminated with UV light it undergoes a rapid uncaging reaction and releases a highly fluorescent red dye.
It can most effectively be excited from 620 to 645 nm. Abberior CAGE 635 is designed for single-molecule photoswitching microscopy modes such as PALM, STORM and GSDIM and, in its uncaged form, also performs well in STED microscopy. It performs very well with Leica GSD microscope and other superresolution microscopes such as Nikon N-Storm.
Further, after photoactivation, Abberior CAGE markers can be tracked in confocal and epifluorescence microscopy to analyses molecular dynamics such as diffusion, flow directions and velocities, e.g. are tracking experiments in which the fluorescence signal is observed over time after the initial release of the fluorophore.
Photophysical properties (carboxylic acid):
Absorption Maximum, λex [nm]: 630 (photoactivated, PBS pH 7.4), 316 (non-photoactivated, PBS pH 7.4)
Extinction Coefficient, εmax [M-1cm-1]: 110 000 (photoactivated, PBS pH 7.4)
Photoactivation Wavelength, λ [nm]: 360-440
Fluorescence Maximum, λem [nm]: 653 (photoactivated, PBS pH 7.4)
Recommended STED Wavelength, λ [nm]: 750-800
Fluorescence Quantum Yield, λ: 0,75 (photoactivated, PBS pH 7.4)
Fluorescence Lifetime, τ [ns]: 3,5 (photoactivated, PBS pH 7.4)
