Application

Adenosine 5′-triphosphate disodium salt hydrate has been used:
  • to prepare adenosine triphosphate (ATP) standard solutions to determine ATP levels in various bacterial cultures
  • bone marrow-derived macrophages (BMDM) treatment
  • as a preparation free of contaminating guanine nucleotides to study the binding effects of coatomer β subunit (β-COP)

Biochem/physiol Actions

Adenosine 5′-triphosphate (ATP) is a central component of energy storage and metabolism in vivo. ATP is used in many cellular processes, respiration, biosynthetic reactions, active transport, and motility. ATP is a substrate of many kinases involved in cell signaling and of adenylate cyclase that produce the secondary messenger cyclic adenosine monophosphate (cAMP). ATP provides the metabolic energy to drive metabolic pumps. It serves as a coenzyme in a wide array of enzymatic reactions.
P2 purinergic agonist; increases activity of Ca2+-activated K+ channels; substrate for ATP-dependent enzyme systems

The coatomer is a cytosolic protein complex that reversibly associates with Golgi membranes and is implicated in modulating Golgi membrane transport. The association of beta-COP, a component of coatomer, with Golgi membranes is enhanced by guanosine 5'-[gamma-thio]triphosphate (GTP[gamma S]), a

Separation of AMP (adenosine monophosphate); ATP (adenosine triphosphate)

Learn about the four membrane-bound protein complexes that make up the electron transport chain metabolic pathway supplying energy as ATP for cellular respiration.

Protocols

To measure luciferase activity, an assay that measures light emission with a luminometer is used. Firefly luciferase is used for detection and quantitation of ATP and as a reporter for genetic function.

To measure hexokinase activity, a continuous spectrophotometric rate-determination assay is used at 340 nm. Hexokinase catalyzes the phosphorylation of D-hexose sugars using ATP as the phosphate source.
