General description

Atg12 is an autophagy-related protein. It forms a complex with Atg5 and functions in LC3 processing.
The autophagy-related protein 3 (ATG3) gene is mapped to human chromosome 5q22.3. ATG12 is a mammalian homolog of yeast Apg3p/ Atg3 is ubiquitously expressed in human tissues.

Immunogen

synthetic peptide corresponding to amino acids 27-39 of mouse Atg12, conjugated to KLH. The corresponding sequence differs by one amino acid in human.

Biochem/physiol Actions

Macroautophagy, usually referred to as autophagy, is a major pathway for bulk degradation of cytoplasmic constituents and organelles. Atg12 is a ubiquitin-like modifier essential for autophagy. Two ubiquitin-like conjugation systems are involved in autophagosome formation: Atg12 and Atg8 conjugation systems. Autophagy is required for normal turnover of cellular components during starvation. It plays an essential role in cellular differentiation, cell death and aging. Defective autophagy may contribute to certain human diseases such as cancer, neurodegenerative diseases, muscular disorders and pathogen infections.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.
