Application

Anode buffer for Tricine gels is to be used in conjunction with Tris-tricine-SDS for SDS – polyacrylamide gel electrophoresis of proteins using the Schagger and von Jagow method. The Schagger and von Jagow method is designed for the separation of small molecular weight proteins. The Schagger and von Jagow method differs from the Laemmli method in that the glycine is replaced with tricine in the cathode buffer and the gel contains 1 M Tris-HCl, pH 8.45 instead of 0.375 M Tris-HCl, pH 8.9. Dilution of the 10 × Anode buffer produces a 1 × buffer containing 0.2 M Tris-HCl, pH 8.9. Recommended running conditions is 125 volts.

Analysis Note

Tested for use as an anode buffer in a tricine gel (See Sigma Technical Bulletin MWM-100).
