General description

PURedit® Cas9 Precision Cytosine Base Editor has been engineered to achieve defined nucleotide changes of cytosine to thymine using RNP complex delivery. The PURedit® Cas9 Precision Cytosine Base Editor will edit cytosine residues from the third position of the protospacer to the ninth position. The base editor protein is recombinantly expressed in E. coli with 2X NLS. Uracil glycosylase inhibitor (UGI) protein, also recombinantly expressed in E. coli with 2X NLS, is included as a recommended optional component to enhance the rate of C-to-T editing and reduce the rate of other editing outcomes, including insertions and deletions (indels). UGI may be omitted to enable lower-frequency C-to-R (R = A or G) editing.

The kit contains:

Cytosine Base Editor protein (CAS9CBEPRC) in 50% glycerol at 8 mg/mL

Uracil glycosylase inhibitor (UGIPRT) in 50% glycerol at 15 mg/mL

Transfection buffer (PEXBUF)

Dilution buffer (DBUFFER)
