CAR-2/3F4, also known as 2/3 F4, was formed by fusing P3X63Ag8-653 mouse myeloma cells with spleen cells from a BALB/c mouse immunised six times with 20μg human thioredoxin (TRX) at monthly intervals. Human TRX was cross-linked to Keyhole Limpet Haemocyanin for this purpose. The antibody is directed against monomeric and polymeric forms of human TRX and shows no cross-reactivity to bacterial TRX or human glutaredoxin. CAR-2/3F4 has been applied as capture antibody in a chemiluminescent detection assay for free human TRX. CAR-XP10 (Sigma Catalogue number. 97011601) can be used as second antibody in this assay.
Preparation Note
Maintain cultures between 2-9 x 100,000 cells / ml; 5% CO2; 37°C; start culture on mouse macrophage feeder layer
RPMI 1640 + 2mM glutamine + 1mM Sodium Pyruvate (NaP) + 15% Foetal Bovine Serum (FBS)
