Application
MC3T3-E1 Cell Line has been used to study and propose an actin filament cytoskeleton analysis framework. It has also been used to study the effect of biodegradable magnesium and magnesium alloys on selected properties of these cells stimulated by direct cell/material interaction.
Biochem/physiol Actions
Mouse C57BL/6 calvaria
The osteoblastic cell line MC3T3-E1 has been established from a C57BL/6 mouse calvaria and selected on the basis of high alkaline phosphatase (ALP) activity in the resting state. Cells have the capacity to differentiate into osteoblasts and osteocytes and have been demonstrated to form calcified bone tissue in vitro. Mineral deposits have been identified as hydroxyapatite. Expression of basic fibroblast growth factor (bFGF) mRNA and protein has been shown to be regulated by treatment with TGF β and bFGF. Prostaglandin F2a has been reported to stimulate DNA synthesis and proliferation by up-regulation of insulin-like growth factor I receptors. MC3T3-E1 secrete collagen and express murine leukemia inhibitory factor (mLIF) in RNA.
Preparation Note
MEM α + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Split sub-confluent cultures (70-80%) 1:4 using 0.25% trypsin or trypsin/EDTA 5% CO2 37ºC.
