General description

Endothelial Cell Growth Supplement (ECGS) from bovine neural tissue is a biologically derived extract containing growth‑promoting factors that support the growth of vascular endothelial cells of mammalian origin. According to reported literature, this supplement comprises acidic fibroblast growth factor (aFGF) and basic fibroblast growth factor (bFGF). ECGS is supplied as a powder and is suitable for use in in vitro mammalian cell culture research.

Application

Endothelial Cell Growth Supplement (ECGS) from bovine neural tissue has been used as a supplement to basal culture media in endothelial research applications, including:

  • Dulbecco’s Modified Eagle Medium (DMEM) supplementation
  • F‑12K medium supplementation
  • Human umbilical vein endothelial cell (HUVEC) culture

Biochem/physiol Actions

Endothelial Cell Growth Supplement (ECGS) has been reported to support the proliferation of human endothelial cells, including human umbilical vein endothelial cells (HUVECs) and human saphenous vein endothelial cells (HSVECs), in vitro. ECGS has been studied as a coordinating factor in endothelial‑related cellular processes, including interactions with cytokine‑mediated pathways and extracellular matrix remodeling in vascular smooth muscle cell models. It has also been reported as a supportive media supplement for hybridoma cell fusion and growth in monoclonal antibody research workflows.

Preparation Note

ECGS is an extract of bovine neural tissue containing growth promoting factors for vascular endothelial cells of mammalian origin. Endothelial cell growth supplement is prepared using a modification of the method of Maciag, et al., and is lyophilized from a sterile solution containing NaCl and streptomycin sulfate.

Analysis Note

The proliferative activity of endothelial cell growth supplement (ECGS) is tested in culture using fetal bovine heart endothelial cells (ATCC CRL 1395) seeded at low density. The EC50 is defined as the effective concentration of growth factor that elicits a 50% increase in cell growth in a cell based bioassay.
