Biochem/physiol Actions

Flotillin 1 is directly connected to the plasma membrane. The association of flotillin 1 depends on the palmitoylation in a conserved Cys34 residue, whereas the association of flotillin 2 depends on both myristolation and palmitoylation. Both the form possess a complementary tissue distribution. Flotillin 1 is mainly expressed in striated muscle tissues, adipose tissue and lung, whereas flotillin 2 is much widely found but absent from skeletal muscle. It has been reported that flotillin 1 act as a molecular link between lipid rafts of the plasma membrane and a multimeric signaling complex at the actin cytoskeleton.

General description

Flotillins are lipid raft-associated proteins belonging to the integral membrane proteins. It is associated in different biological processes such as endocytosis, phagocytosis, neuronal regeneration, insulin signaling etc. It exhibits an evolutionarily conserved domain called the prohibitin homology (PHB) domain or SPFH domain (stomatin/prohibitin/flotillin/HflK/C domain). The lipid recognition motif present in the PHB domain, is responsible for targeting proteins to the plasma membrane. Flotillin protein mainly possess two forms, flotillin 1 and 2. They are also known as reggie-2 and 1, respectively.

Immunogen

synthetic peptide corresponding to amino acid residues 413-428 of mouse flotillin 1, conjugated to KLH. The corresponding sequence is identical in rat and differs by one amino acid in human.

Physical form

Solution in 0.01 M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.

Flotillin-1 is a lipid raft-associated protein that has been implicated in various cellular processes. We examined the subcellular distribution of flotillin-1 in different cell types and found that localization is cell type-specific. Flotillin-1 relocates from a cytoplasmic compartment to the
