Immunogen
α-2-Macroglobulin (validated) native human protein.
Immunogen Sequence: GI # 68765, sequence 1-1474
Application
Anti-α-2-Macroglobulin antibody produced in chicken is suitable for western blotting at a working dilution of 1:500 and for cell staining at a working dilution of 1:200. It is also used for indirect ELISA.
Biochem/physiol Actions
A2M (α-2-macroglobulin) inhibits all four classes of proteinases by a unique ′trapping′ mechanism. This protein undergoes a conformational change when a proteinase binds and cleaves a specific peptide stretch called the "bait region" in the protein. This modified protein traps the bound proteinase It is involved in the clearance and degradation of Aβ, which is a major component of beta-amyloid deposits. Therefore, it is associated with Alzheimer′s disease (AD).
Physical form
Solution in phosphate buffered saline containing 0.02% sodium azide.

Circular dichroism spectroscopy, sedimentation velocity and ultraviolet difference spectroscopy were used to compare alpha 2-macroglobulin, alpha 2-macroglobulin-trypsin complex and alpha 2-macroglobulin-methylamine complex. The circular dichroic spectrum of native alpha 2-macroglobulin is significantly changed in shape and magnitude following reaction with

Alzheimer's disease is a rapidly worsening public health problem. The current lack of effective treatments for Alzheimer's disease makes it imperative to find new pharmacotherapies. At present, the treatment of symptoms includes use of acetylcholinesterase inhibitors, which enhance acetylcholine levels

Glucose regulated protein 78 (GRP78) is expressed on cell surface in exceptional conditions as seen in cancer cells and macrophages. We have reported its membrane localization in sperm. The functional significance of its surface localization in sperm is an enigma.
