General description

Biotin phosphoramidite is a reagent used to directly incorporate a biotin label at the 5′ hydroxy group of the terminal nucleoside base through a p-aminophenethyl spacer group during oligonucleotide synthesis.

The photocleavable biotin phosphoramidite reagent is compatible with automated DNA/RNA synthesizers using phosphoramidite chemistry. The length of the p-aminophenethyl spacer group allows streptavidin affinity purification of the oligonucleotide by reverse-phase HPLC. Biotinlabeling is compatible with PCR and most hybridization techniques.Key Features of Biotin Phosphoramidite
  • Behaves like any standard phosphoramidite on the DNA synthesizer
  • Readily soluble in acetonitrile
  • Biotin compound has a dimethoxytrityl (DMT) group on the ring structure,which allows coupling-yield determination and trityl-selective purification• A long, water-compatible linker arm ensures maximum sensitivity
  • Provides a coupling efficiency of at least 95%The product is configured for Expedite and PolyGen® Synthesizers.

Application

Biotin Phosphoramidite is suitable for the synthesis of 5′-biotinylated oligonucleotides whose applications are seen in ligase-mediated gene detection, direct dideoxy sequencing following PCR, and non-radioactive sequencing of DNA.
