General description

α−Mannosidase from Canavalia ensiformis is a tretamer composed of two subunits that each contain two components at 44 and 66 kDa.
α-Mannosidase is an exo-glycosidase and an inc metalloenzyme.
Lysosomal α-mannosidases are class II mannosidases that belong to glycoside hydrolase family 38 and play an important role in the degradation of asparagine-linked carbohydrates of glycoproteins.

Application

α-Mannosidase can be used to liberate mannose from a variety of synthetic and natural α-mannosides. It has also been used in a study to investigate the causes of neurodegeneration in mucolipidosis II ′knock-in′ mice.
α-Mannosidase from Canavalia ensiformis (Jack bean) has been used:
  • for the digestion of neutral glycans.
  • to remove O-glycans from recombinant β-glucosidase
  • for the inhibition of renal fibroblasts proliferation

Liberates mannose from a variety of synthetic and natural α-mannosides.

Biochem/physiol Actions

α−Mannosidase has been shown to inhibit the proliferation of B-lymphocytes.
α-Mannosidase cleaves near α1−2-, α1−3- and α1−6-linked mannose residues.
α-Mannosidase is an acid hydrolase which is located in plant vacuoles and is thought to be involved with the turnover of N-linked glycoproteins.

Unit Definition

One unit will hydrolyze 1.0 μmole of p-nitrophenyl α-D-mannoside to p-nitrophenol and D-mannose per min at pH 4.5 at 25°C.
