Application

Organo/Limonene Mount is a ready-to-use mounting medium is made with limonene, a natural product from orange peels. It is good for coverslipping stained tissues and cell smears that can be dehydrated with organic solvents. This mounting medium is suitable for organic solvent resistant immunohistochemistry (IHC) chromogens as well as an excellent choice for mounting H and E stained slides.

  1. How does the storage temperature relate to shipping conditions?

    The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.   6. What are the instructions on preparing a sample for use with Product No. O8015 (Organo/Limonene Mount)?

    Dehydrate slides with dehydrating agents, such as graded ethanol or reagent alcohol. Dehydration refers to removing the water by using a series of grades alcohols. Typically slides spend 1-5 minutes each in a series of alcohols. Sequences vary by laboratory. A typical sequence would be 1 station of 80% alcohol, 2-3 changes of 95% alcohol, 2-3 changes of absolute alcohol. The slides are then processed through 2-3 changes of xylene. After finishing with the last xylene solution, the slides are ready for coverslipping with the Organo/Limonene Mount.

  7. The instructions for use of Product No. O8015, Organo/Limonene Mount, call for using xylene for clearing.  Can limonene or other xylene substitutes be considered acceptable for use?

    Yes, limonene would be considered a suitable xylene substitute. There are however a wide variety of xylene substitutes available on the market. No exhaustive comparative studies have been performed using Organo/Limonene Mount with each and every available xylene substitute.

  8. Organo/Limonene Mount (Product No. O8015) is stated to be suitable for use with chromogens for immunohistochemistry.  Does this?

    Suitable chromogens for immunohistochemistry would be DAB, or DAB with an added metal such as nickel or cobalt.  Alkaline phosphatase chromogens would also be suitable for use with Oregano/Limonene Mount.

  9. Why is Organo/Limonene Mount (Product No. O8015) preferable to a mounting media that is xylene based?

    There are more health hazards associated with xylene in comparison to products derived from natural products, such as Organo/Limonene Mount.
