Application

Pyruvate dehydrogenase contributes to linking the glycolysis metabolic pathway to the citric acid cycle and releases energy via NADH. It is used to study how aerobic training effects pyruvate dehydrogenase . It is also used to study the interaction between pyruvate dehydrogenase kinase and the pyruvate dehydrogenase complex as well as the function and regulation of the pyruvate dehydrogenase complex. Product P7032 is provided in a buffered aqueous glycerol solution and is from porcine heart.
The enzyme has been used in a primary enzyme assay to test the ability of certain compounds to reduce PDK- (pyruvate dehydrogenase kinase) catalyzed PDC (pyruvate dehydrogenase complex) inactivation.

Biochem/physiol Actions

Pyruvate dehydrogenase is involved in the decarboxylation of pyruvate and reductive acetylation of lipoic acid within the pyruvate dehydrogenase complex (PDC). Pyruvate dehydrogenase kinase (PDK) inactivates pyruvate dehydrogenase while pyruvate dehydrogenase phosphatase activates pyruvate dehydrogenase.

Physical form

Supplied as a 50% glycerol solution containing ~9 mg/mL bovine serum albumin, 30% sucrose, 1.5 mM EDTA, 1.5 mM EGTA, 1.5 mM 2-mercaptoethanol, 0.3 TRITON® X-100, 0.003% sodium azide, and 15 mM potassium phosphate, pH 6.8

Other Notes

One unit will convert 1.0 μmole of β-NAD to β-NADH per min at pH 7.4 at 30 °C in the presence of saturating levels of Coenzyme A.
