General description

The protein encoded by this gene is a member of the kelch family of proteins, which is characterized by a 44-56 amino acid repeat motif. The kelch motif appears in many different polypeptide contexts and contains multiple potential protein-protein contact sites. Members of this family are present both throughout the cell and extracellularly, with diverse activities. (provided by RefSeq)

Biochem/physiol Actions

KLHL20 (kelch like family member 20) is a substrate adaptor of cullin3 (Cul3). It is required for post-Golgi trafficking. The Cul3-KLHL20 complex works as an ubiquitin E3 ligase and is responsible for polyubiquitination of coronin-7, thereby targeting coronin-7 to trans-Golgi network. KLHL20 is also involved in autophagy by regulating autophagy-associated degradation of ULK1 (unc-51 like autophagy activating kinase 1) and VPS34 (vacuolar protein sorting 34) complex subunits. Additionally, KLHL20 is associated with hypoxia responses and tumorigenesis. HIF1 (hypoxia inducible factor 1)-mediated upregulation of KLHL20 gene causes hypoxia-induced PML (promyelocytic leukemia) polyubiquitination. PML destruction is linked with epithelial-mesenchymal transition, tumor growth, angiogenesis and other related events.
