Biochem/physiol Actions

SFRP1 (secreted frizzled-related protein 1) plays a major role in the progression of several malignancies. It also plays a vital role in antagonizing Wnt signaling. Overexpression of SFRP1 is linked with apoptosis.
Secreted Frizzled-related protein 1 (SFRP1) is a secreted inhibitor of Wnt signaling pathway. DNA damage or oxidative stress causes cellular senescence, which leads to the overexpression of sfrp1. This protein induces senescence phenotypes via the inactivation of Wnt signaling and activation of the retinoblastoma (Rb) pathway. Mutations or aberrant methylation in this gene impair the induction of senescence in several cancers, including breast cancer, bladder cancer, and colorectal tumorigenesis.

General description

SFRP1 (secreted frizzled-related protein 1) is a 35.4 kDa antagonist protein.
The encoded protein by SFRP1 gene has a cysteine-rich domain similar to the WNT-binding site of Frizzled receptors. It belongs to the WNT pathway. This gene is located on human chromosome 8p12–11.1.
The gene SFRP1 (secreted frizzled related protein 1) is mapped to human chromosome 8p22–p12, a region frequently deleted in breast cancer. It is highly expressed in normal breast tissue.
