General description
Neurotrophic receptor tyrosine kinase 3 (NTRK3), also known as tropomyosin receptor kinase C (TRKC), with 20 exons and 19 introns, is mapped to human chromosome 15q25. NTRK3 encodes the high-affinity receptor for neurotrophin 3 (NT-3). The encoded 145kDa glycoprotein belongs to tropomyosin-related kinase (Trk) family of neurotrophin receptors. NTRK3 is characterized with a signal sequence, two cysteine-rich domains flanking a leucine-rich domain, two Ig-like domains of the C2 type, a transmembrane domain and a tyrosine kinase domain.

Biochem/physiol Actions
Neurotrophic receptor tyrosine kinase 3 (NTRK3) plays a vital role in regulation of cell proliferation, survival and differentiation in the mammalian nervous system, especially those of sensory neurons involved in proprioception. It is also involved in the formation of the mammalian heart. TRKC facilitates initiation, progression and metastasis of many tumors in humans. TRKC is overexpressed in neuroblastomas as well as medulloblastomas and also in various non-neuronal tissues including medullary thyroid carcinoma, breast cancer, colon cancer, lung cancer, liver cancer, prostate cancer. The encoded protein acts as a receptor for the neurotrophin 3′-nucleotidase (NT3), which is involved in oligodendrocyte and myelin formation. Variation in the gene affects white matter integrity in brain regions associated with neuropsychiatric disorders, including major depressive disorder (MDD-RE). TRKC stimulates the expression of serine/threonine-protein kinase PLK-1 and Twist-1 transcription factor via activation of Akt (a serine/threonine kinase also known as protein kinase B [PKB]) / mammalian (or mechanistic) target of rapamycin (mTOR) pathway and inhibits apoptosis. Thus, it facilitates leukemogenesis.
