Rilpivirine inhibits the activity of the reverse transcriptase enzyme, which is essential for the replication of HIV-1. Specifically, it binds to a specific site on the enzyme and blocks its activity, thereby preventing the conversion of viral RNA into DNA, which is an essential step for the integration of viral DNA into host cells.
Rilpivirine is a next generation nonnucleoside reverse transcriptase inhibitor (NNRTI), It has been shown to be active against many HIV strains with mutations that confer resistance to the first-generation NNRTIs.
