XAP044 is a brain penetrant, potent and selective mGlu7 (metabotropic glutamate receptor subtype 7) antagonist that inhibits lateral amygdala long-term potentiation (LTP) in brain slices from wild type mice. XAP044 binds to a pocket localized in mGlu7′s extracellular Venus flytrap domain (VFTD) close to the binding site of the orthosteric agonist L-glutamate. XAP044 exhibits a wide-spectrum anti-stress, antidepressant- and anxiolytic-like efficacy in rodent behavioral model.
XAP044 is a brain penetrant, potent and selective mGlu7 antagonist.
