BAY-294 corresponds to the (S)-enantiomer of and the recommended control for the selective SOS1-KRAS interaction inhibitor BAY-293 (IC50 = 2.34 μM/BAY-294 vs. 21 nM/BAY-293; 10 nM hSOS1cat & 5 nM hKRAS G12C). BAY-294 exhibits little potency against SOS1-mediated KRAS activation (IC50 = 3.91 μM/BAY-294 vs. 52 nM/BAY-293; 10 nM SOS1cat & 50 nM wt hKRAS).
