2′-Deoxyadenosine 5′-diphosphoribose (2′- deoxy-ADPR) is a superagonist of the TRPM2 channel (transient receptor potential melastatin 2), a Ca2+-permeable, voltage independent, nonselective cation channel implicated in free-radical induced cell death, chemotaxis, insulin secretion, thermosensation, immune cell responses and activation of the inflammasome. 2′-Deoxy-ADPR caused 10.4-fold higher whole-cell currents at saturation than adenosine diphosphoribose (ADPR), which had been previously considered to be the main intracellular second messenger that activated TRPM2. 2′-Deoxy-ADPR decreased the inactivation rate resulting in higher average open probability.
