Santacruzamate A (CAY10683) is a cell-permeable and bioavailable carbamate compound that potently inhibits HDAC2 (IC50 = 119 nM) selectively over HDAC4 and HDAC6 (IC50 = > 1 μM and 433.5 nM, respectively). CAY10683 readily enhances cellular acetylated HDAC2 levels upon LPS-stimulation and only modestly inhibits HuT-78 cutaneous T-cell lymphoma and HCT-116 colon carcinoma cells growth (GI50 = 1.4 and 29.4 uM, respectively). It alleviates acute liver failure (ALF) in rats (i.p.) and rescues cognitive deficits in APPswe/PS1dE9 mice (p.o.).
