Selective α-synuclein (α-syn) translation inhibitor with cytoprotective efficacy against preformed α-syn fibrils (PFFs) in SH-SY5Y neuronal cultures.
Synucleozid is a selective α-synuclein (α-syn) translation inhibitor (46% & 69% α-syn downregulation post 48 h, respectively, with 0.5 or 1 μM Synucleozid) with cytoprotective efficacy against preformed α-synuclein fibrils (PFFs) in SH-SY5Y neuronal cultures (56% protection; 0.5 μM Synucleozid added 24 h prior to 48 h 50 μg/mL PFFs exposure). Synucleozid directly and selectively targets α-syn (SNCA) mRNA 5′ UTR at the iron-responsive element (IRE) site, effectively interfering with the polysomes assembly onto SNCA mRNA.
