Hypoxia probe 4 is a Tokyo green-based non-fluorescent hypoxia probe that undergoes cytochrome P450 reductase (PH51)-catalyzed reduction to yield the fluorescent reporter Tokyo green (ex 480 nm, em 515 nm) in conditions of 0.5% O2 and lower, allowing imaging hypoxia levels similar to those detected by EF5 without the use of antibodies. When employed together with resorufin-based hypoxia probe 2 (ex 545 nm, em 582 nm), the probe pair allows the simultaneous (two-color) imaging of different hypoxia levels in living cells.
