Potent heat shock protein 70 inducer with therapeutic efficacy in a rat model of transient cerebral ischemia in vivo.
TRC051384 is a potent heat shock protein 70 inducer (fold of HSP70 mRNA induction in 4h at 6.25/12.5 µM = 19/210 in HeLa & 10/84 in primary neonatal rat cerebellum neurons) that increases HSF1 transcriptional activity in HeLa cells (by 2.1/3.3-fold at 12.5/2.5 µM) and reduces LPS-induced TNF-α expression in THP-1 cells (% inhibition/conc. = 60/6.25 µM & 90/12.5 µM). TRC051384 reduces neuronal injury and disability in a rat model of transient cerebral ischemia in vivo (9 mg/kg 4h or 8h post onset of ischemia, then 4.5 mg/kg every 2h for 48h via i.p.).
