BAY-593, a GGTase-I inhibitor, primarily targets the YAP1/TAZ signaling pathway, a crucial axis in cell proliferation and tumor progression. Notably, BAY593 alters the mRNA expression levels of YAP1/TAZ target genes including CYR61, CTCF, ANKRD1, and CD274, indicating its specific molecular action. In vitro studies reveal its IC50 values: 9.41 nM for MDA-MB-231 TEAD-luc, 0.564 µM for MDA-MB-231 proliferation, and 38.4 nM for HT-1080 proliferation. The efficacious in vivo dose was determined as 5 mg/kg (p.o., BID), in the PXF 541 pleuromesothelioma xenograft model, showcasing its therapeutic potential in cancer treatment.
