Survivin expression inhibitor with anti-cancer efficacy in cultures and in vivo.
YM155 is a survivin expression inhibitor (effective conc. 10-100 nM for 6-48h; PC-3 & PCC-1 cells) that induces apoptosis in malignant melanoma and hormone-refractory prostate cancer (HRPC) cultures (GI50 = 2.3-11 nM) by targeting survivin gene promoter activity without affecting the expression of other IAPs or Bcl-2-related proteins even at concentrations as high as 100 nM. In a s.c. xenografted PC-3 tumor model in mice, 3-day continuous infusions of YM155 at 3 to 10 mg/kg induced massive tumor regression in vivo.
