Ynamide-2e, featuring a N-trifyl group, provides a highly efficient and chemoselective method for cysteine-specific modification in peptides and proteins. Ynamide-2e exclusively forms Z-isomer conjugates with superior stability under various conditions. This bioconjugation tool exhibits excellent thiol specificity, with a second-order rate constant of 0.2399 M−1 s−1 for glutathione, outperforming traditional reagents like iodoacetamide in reducing off-target labeling. Functioning efficiently in aqueous buffer, Ynamide-2e is compatible with a wide range of proteins, including antibodies, without compromising protein integrity or function. Its applicability in bottom-up proteomics workflows and potential for redox proteomics make it valuable for studying protein modifications and interactions.
