6PPD-quinone (6PPDQ), a highly toxic tire antioxidant 6PPD derivative, bioaccumulates in lipid-rich tissues. 6PPD-Q primarily disrupts vascular permeability (potentially the blood-brain barrier) and induces mitochondrial dysfunction, causing oxidative stress. Molecularly, 6-PPDQ targets cannabinoid receptors (notably high-affinity CNR2), affects neuronal glycolysis, TCA intermediates, and promotes α-synuclein aggregation in vitro. Cellular/biological outcomes include dose-dependent mammalian multi-organ toxicity (hepatic, enteric, pulmonary, neuro-, reproductive [PCOS-linked], cardiac). In vivo, its extreme relevance is evident in potent salmonid lethality (e.g., Coho salmon LC50: 0.041 µg/L).
