Aminophenyl-fluorescein (APF), a cell-permeable and photostable probe, is designed for the selective detection of highly reactive oxygen and nitrogen species (hROS & hRNS), notably hypochlorite (OClˉ), hydroxyl radicals (HO•), and peroxynitrite (ONOOˉ) over minimally reactive H2O2, O2ˉ•, NO, 1O2 and ROO•. Initially exhibiting weak fluorescence (ε455 = 24,000 M-1cm-1, Φ: 0.008), APF undergoes O-dearylation upon reaction with these species, releasing its amino-phenoxy group and generating intensely fluorescent fluorescein, which leads to a dose-dependent fluorescence signal increase (λex/λem: 490/515 nm) and significantly enhanced optical properties (ε492 = 84,000 M-1cm-1, Φ: 0.85). APF′s inherent selectivity, complete autoxidation resistance, low toxicity, and quantitative fluorescence response establish it as a valuable tool for applications spanning microscopy, enzymatic assays, and multiplexing.
