HADA, a non-toxic, blue-emitting (λex/λem: 405/450 nm) 7-hydroxycoumarin-labeled D-amino acid analog, covalently incorporates into bacterial peptidoglycan (PG) via periplasmic transpeptidases, primarily at D-alanine residues, enabling real-time visualization of PG dynamics, spatiotemporal growth, and cell wall synthesis across diverse bacteria. This highly versatile and specific live cell probe with moderate photostability facilitates multiplexed imaging and antibiotic and microbiome studies, utilizing bioorthogonal approaches for rapid, covalent PG labeling across a broad taxonomic range and permitting in situ studies.
