Tildipirosin

Tildipirosin, a cell-permeable macrolide antibiotic, shows enhanced efficacy against key Gram-negative respiratory pathogens like Mannheimia haemolytica and Pasteurella multocida through improved outer membrane penetration. Selectively inhibits bacterial protein synthesis (IC50 ~0.23 µM) by binding the 23S rRNA of the 50S bacterial ribosomal subunit, blocking peptide chain prolongation and disrupting vital functions, yielding bacteriostatic or bactericidal effects with typical MIC90 values of 0.5-4 µg/ml. This mechanism enables effective treatment and metaphylaxis of Bovine (BRD) and Swine Respiratory Diseases (SRD), often as a single dose due to its long duration of action. Tildipirosin also displays immunomodulatory and anti-inflammatory actions by inhibiting pro-inflammatory cytokine secretion. Its significance is providing a targeted, long-acting therapeutic for critical veterinary respiratory infections.
