SALAD1 (structure-informed, array-enabled LigAnD 1) is a cell-permeable, photostable, and ultrabright RNA-activated fluorophore. SALAD1 specifically and reversibly targets the Mango II aptamer (KDapp = 0.69 nM) with high selectivity. Upon binding, SALAD1 shows a remarkable 1,500-fold increase in fluorescence (λex 511 nm, λem 540 nm; Φ = 0.722, ε = 47,284 M⁻¹ cm⁻¹). This enables superior real-time confocal RNA imaging within living cells. Furthermore, the Mango III–SALAD1 complex, while binding less tightly, offers even greater brightness, expanding its potential for diverse live-cell imaging applications.
