Biotin-LC-Tyramide is a bifunctional probe, featuring a phenol moiety for enzymatic oxidation and a biotin group for high-affinity streptavidin-based detection, engineered for high-fidelity proteomic mapping and signal amplification. Its core utility stems from an optimized 6-atom "LC" (Long Chain) aminohexanoic acid spacer, extending ~20.3 Å to position the biotin reporter away from the target protein surface, thereby mitigating steric hindrance for superior streptavidin binding. Activated by peroxidases, Biotin-LC-Tyramide generates a short-lived radical that covalently labels proximal tyrosine residues with high spatial resolution. This mechanism enables potent tyramide signal amplification (TSA) for high-resolution mapping in immunohistochemistry (IHC) and fluorescent in-situ hybridization and is a key reagent for proximity labeling techniques like APEX-MS to identify organelle-specific proteomes and protein-protein interactions in living cells.
