Crucially, its elevated reactivity dictates a tighter effective capture radius (<10 nm effective capture) via rapid radical quenching and self-polymerization, thereby eliminating the broad "diffusion cloud" and background noise associated with traditional Biotin-Phenol (BP1). This physiochemical refinement prioritizes the labeling of direct interactors over bystanders, enabling the high-fidelity capture of weak and transient assemblies often lost during cell lysis. Validated against highly dynamic signaling networks—including EGFR adaptors (GRB2, STS1) and the ILK complex—BP5 successfully differentiates interactions indistinguishable by BP1, providing a robust platform for mapping mobile interactomes and protein translocation events at organelle interfaces.
