General description

Thymidine is also referred to as pyrimidine deoxynucleoside. Deoxythymidine is a nucleoside present in DNA. In a DNA double stranded structure, thymidine pairs with deoxyadeninosine.

Application

Thymidine can be used in HT and HAT cocktails for hybridoma fusion, selection and cloning. It has been used:
  • as a nucleotide source in minimal essential medium to evaluate the outgrowth of the iSCNT-derived blastocysts
  • as a supplement in Dulbecco′s Modified Eagle Medium (DMEM) selection medium used for the production of anti-neurogranin (NRGN) antibodies
  • to prepare THMG (thymidine, hypoxanthine, glycine, methotrexate) and THG stock solution for cleansing mouse lymphoma cells

Biochem/physiol Actions

Thymidine is useful in cell synchronization during S-phase. In the salvage pathway of pyrimidines, pyrimidine phosphorylase reversibly converts thymine to thymidine.

    This product is soluble in water (50 mg/mL), yielding a clear, colorless solution with heating. For aqueous concentrations higher than 50 mg/mL, solubility can be enhanced with the addition of a small amount of NaOH solution. Add the base drop wise with slow stirring. Thymidine is also soluble in methanol, hot alcohol, hot acetone, hot ethyl acetate, pyridine, glacial acetic acid, and is sparingly soluble in hot chloroform.
