General description
ThermaStop™ is a reversible hot-start reagent that is compatible with thermostable DNA polymerases. It acts directly on the polymerase to prevent non-specific enzymatic activity below 50°C. Polymerase activity is fully restored at 60°C, but is inhibited again by ThermaStop™ by cooling the reaction. Adding ThermaStop™ to your PCR reactions improves sensitivity and total product yield.
Application
ThermaStop™ has been used as a component of the PCR mix for the human immunodeficiency virus (HIV-1) proviral genome amplification by non-multiplexed nested PCR. It has also been used for the amplification of DNA from oral swabs of HIV infected individuals.
Features and Benefits
- Adding ThermaStop™ to your PCR reactions improves sensitivity and total product yield
- Increases the product yield for downstream applications like genotyping, next-generation sequencing (NGS) sample preparation, Gibson assembly, diagnostics, and multiplex PCR
- It improves polymerase specificity, enhances PCR performance and quantitative data
- Prevents primer-dimer formation and non-specific products before PCR
- Enables stabilization of master mixes at room temperature before PCR for up to 36 hrs
- Rapidly inactivated during the first PCR cycle
- Rapidly re-activated upon cooling
Preparation Note
Dissolve in Tris pH 8.3 buffer

Clonal expansion of HIV-infected cells contributes to the long-term persistence of the HIV reservoir in ART-suppressed individuals. However, the contribution from cell clones that harbor inducible proviruses to plasma viremia is poorly understood. Here, we describe a single-cell approach to
