General description

The REDExtract-N-Amp Tissue PCR Kit contains all the reagents needed to rapidly extract and amplify genomic DNA from mouse tails and other animal tissues, buccal swabs, hair shafts, and saliva. There is no need for mechanical disruption, organic extraction, column purification, or precipitation of the DNA. The kit includes a PCR ReadyMix, specially formulated for direct PCR amplification from the extract. This formulation uses an antibody-based hot start, for specific amplification. The REDExtract-N-Amp Tissue PCR Kit contains a tracking dye that allows for convenient direct loading of PCR reactions onto agarose gels for analysis.

The REDExtract-N-Amp Tissue PCR Kits provide all the reagents necessary to rapidly extract DNA from a wide variety of cells and tissues and amplify targets of interest by PCR. A novel extraction method eliminates the need for long enzymatic digestions or homogenization. The kit also includes a specially formulated hot start PCR reaction mix for amplification directly from the extract. The REDExtract-N-Amp PCR ReadyMix contains an inert dye that acts as a tracking dye and allows for convenient loading of PCR reactions onto agarose gels for analysis.

Principle

The kit comes with validated protocols to extract and amplify genomic DNA from mouse-tails, hair, animal tissue, saliva, and buccal swabs. In a typical procedure, genomic DNA is extracted from a sample that has been incubated in the tissue preparation solution and extraction solution for 10 minutes at room temperature. The sample is heated to 95 °C for 3 minutes and then mixed with a third solution to neutralize inhibitory substances prior to PCR. A portion of the DNA extract is then added to a PCR reaction containing primers and REDExtract-N-Amp PCR ReadyMix, included in the kit.

  1. How do I quantitate my DNA with XNAT, REDExtract-N-Amp® Tissue PCR Kit?

    The DNA extract generated using the Extract-N-Amp kit is very crude, and it is not possible to obtain a reliable 260/280 ratio.  For quantitation of pure DNA, we recommend using one of our GenElute genomic DNA kits.

  2. How do I store my DNA samples for use with XNAT, REDExtract-N-Amp®  Tissue PCR Kit?

    DNA samples can be stored for up to 6 months at 2-8°C with the tissue sample removed.

  3. Are the XNAT, REDExtract-N-Amp® Tissue PCR Kit components available separately?

    Only R4775, the REDExtract-N-Amp PCR Reaction Mix is available separately.

  4. What is the shelf life of the kit XNAT, REDExtract-N-Amp® Tissue PCR Kit and how should I store it?

    The shelf life of XNAT has not been determined.  However, based on our storage guidelines, it should be usable for 1 year from the date of receipt. The REDExtract-N-Amp Tissue PCR Kit can be stored at 2-8°C for up to three weeks. For long-term storage, greater than three weeks, -20°C is recommended. Do not store in a "frost-free" freezer.  We recommend to store in 10 reaction aliquots.

  5. I am unable to detect any PCR product with Product XNAT, REDExtract-N-Amp® Tissue PCR Kit.

    From XNAT Technical Bulletin, troubleshooting guide table:  PCR reaction may be inhibited due to contaminants in the tissue extract.  Dilute the tissue extract with a 50:50 mix of extraction and neutralization solutions.  To test for inhibition, include a DNA control and/or spike a known amount of template (100-500 copies) into the PCR along with the tissue extract.
