General description

The ZytoLight® SPEC 1p36/1q25 Dual Color Probe (PL34, Z-2075-50/200) is a FISH probe designed for the qualitative detection of deletions in the human chromosomal region 1p36.31 (ZyOrange-labeled, chr1:5,808,946-6,176,336, 4.5 ng/µl) and the reference sequences on chromosome 1q25.3 (ZyGreen-labeled, chr1:184,271,714-184,986,522, 10.0 ng/µl) in formalin-fixed, paraffin-embedded (FFPE) specimens, including gliomas. This probe is intended to be used in conjunction with the ZytoLight FISH-Tissue Implementation Kit (Prod. No. Z-2028-5/-20).

Principle:

The ZytoLight® system utilizes the fluorescence in situ hybridization (FISH) technique to detect and visualize specific nucleic acid sequences using direct-labeled probes, thereby eliminating the need for fluorophore-coupled antibodies. During the FISH process, fluorescently-labeled DNA fragments co-denature with their complementary target DNA strands, followed by annealing during hybridization. Unbound probe fragments are subsequently removed through washing steps. The hybridized probe fragments are then counterstained with DAPI and visualized using fluorescence microscopy.In this system, orange signals indicate the 1p36 locus, while green signals represent the 1q25 locus. The microscopy setup employs an exciter filter for wavelength-specific excitation, a dichroic mirror for reflection, and a barrier filter to isolate the emitted light. This optimized ZytoLight® system is particularly effective for identifying genetic aberrations, such as deletions, in formalin-fixed, paraffin-embedded (FFPE) specimens.

Probe Composition:

  • ZyOrange: Labeled polynucleotides (excitation 547 nm/emission 572 nm) targeting sequences in 1p36.31 (chr1:5,808,946-6,176,336).
  • ZyGreen: Labeled polynucleotides (excitation 503 nm/emission 528 nm) targeting sequences in 1q25.3 (chr1:184,271,714-184,986,522).
  • Hybridization Buffer: Formamide-based hybridization buffer.

Application

The probe is designed for qualitative detection of 1p36.31 sequences and 1q25.3 specific sequences in formalin-fixed, paraffin-embedded (FFPE) tissue specimens by FISH. It is used in combination with the ZytoLight® FISH-Tissue Implementation Kit for specimen pretreatment, hybridization, and post-hybridization processing.

Analysis Note

In normal interphase cells, two orange signals (representing the 1p36 locus) and two green signals (representing the 1q25 locus) are expected. In cells with deletions affecting the 1p36 locus, one or no orange signal may be observed. Variations in signal patterns may indicate different rearrangements and should be further investigated.
