General description
The ZytoLight® SPEC MDM4/1p12 Dual Color Probe (PL39, Z-2080-200) is a FISH probe designed for the qualitative detection of amplifications involving the human MDM4 gene and specific sequences on chromosome 1p12 in formalin-fixed, paraffin-embedded (FFPE) specimens. This probe utilizes ZyGreen-labeled polynucleotides to detect MDM4 gene amplifications at 1q32.1 (chr1:204,126,022-204,882,307, 10 ng/µl) and ZyOrange-labeled polynucleotides for the 1p12 reference sequences (chr1:119,537,102-119,823,147, 4.5 ng/µl). It is intended for professional use only in certified, licensed anatomic pathology laboratories under the supervision of qualified personnel and is to be used in conjunction with the ZytoLight FISH-Tissue Implementation Kit (Prod. No. Z-2028-5/-20).
Principle:
The ZytoLight® system employs fluorescence in situ hybridization (FISH) to detect and visualize specific nucleic acid sequences in formalin-fixed, paraffin-embedded (FFPE) specimens. Direct-labeled probes co-denature with their complementary target DNA strands, enabling hybridization. Application
ZytoLight® SPEC MDM4/1p12 Dual Color Probe aids differential diagnosis of various cancers via MDM4 (p53 regulator, aka HDMX/MDMX) amplification detection; MDM4 inhibits p53 like MDM2, relevant for antitumor therapies targeting both.
For the qualitative detection of human MDM4 gene amplifications and chromosome 1p12 specific sequences by fluorescence in situ hybridization (FISH)
Analysis Note
In normal interphase nuclei, the expected results are two green signals (for the MDM4 gene) and two orange signals (for the 1p12 locus). Cells with amplification of the MDM4 gene or aneuploidy of chromosome 1 will exhibit multiple copies of the green signal or large green signal clusters. Variations in signal patterns may indicate different genetic rearrangements and should be investigated further.
