Specificity

Clone1112-1 is a ZooMAb® mouse recombinant monoclonal antibody that detects RABV Glycoprotein. It targets an epitope within the Glycoprotein of ERA (Evelyn Rokitniki Abelseth) strain Rabies virus.

Application

Quality Control Testing

Evaluated by Immunocytochemistry in L929 cells transfected with Rabies Virus Glycoprotein.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected RABV Glycoprotein in L929 cells stably transfected with glycoprotein of ERA strain of Rabies virus.

Tested Applications

Flow Cytometry Analysis: 1 µg from a representative lot detected RABV Glycoprotein in L929 cells stably transfected with glycoprotein of ERA strain of Rabies virus.

Neutralizing Assay: A representative lot neutralized Rabies Virus (Bunschoten, H., et. al. (1989). J Gen Virol. 70 ( Pt 2):291-8; Dietzchold, B., et. al. (1992). Target description

Glycoprotein (UniProt: P03524) is encoded by the G gene in Rabies virus. The glycoprotein (G) of the Evelyn Rokitniki Abelseth (ERA) strain of the rabies virus is a single-pass type I membrane protein that plays a critical role in the virus′s ability to infect host cells. It is composed of a single polypeptide chain, featuring an ectodomain, a transmembrane domain, and a short cytoplasmic tail. The ectodomain contains multiple glycosylation sites, which contribute to its stability and immunogenicity. The primary function of the ERA glycoprotein is to mediate viral entry into host cells by binding to nicotinic acetylcholine receptors (nAChRs) on neuronal cells. It facilitates the fusion of the viral envelope with the host cell membrane, allowing the viral genome to enter the host cell. The glycoprotein is crucial for eliciting an immune response. The glycoprotein is expressed during the viral replication cycle, primarily in infected neurons and other susceptible cells. Its expression is regulated by the rabies virus′s life cycle, peaking during the assembly and budding phases of viral replication. The glycoprotein is localized at the plasma membrane of infected cells, where it is incorporated into new virions. As a key component of the rabies virus, the ERA glycoprotein is directly involved in the pathogenesis of rabies. Infection leads to severe neurological symptoms and is often fatal if not treated promptly. The glycoprotein′s immunogenic properties make it a critical target for rabies vaccines, which aim to elicit an effective immune response against the virus. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
