General description

GDP-L-fucose synthase (UniProt: Q13630; also known as EC:1.1.1.271, GDP-4-keto-6-deoxy-D-mannose-3,5-epimerase-4-reductase, Protein FX, Red cell NADP(H)-binding protein, Short-chain dehydrogenase/reductase family 4E member 1) is encoded by the GFUS (also known as SDR4E1, TSTA3) gene (Gene ID: 7264) in human. In mammals, fucosylated glycans play important roles in ABO blood group antigens, host-microbe interactions, and several human diseases, including rheumatoid arthritis, pancreatic cancer, and hepatocellular carcinoma. GDP-L-fucose synthase is a homodimeric protein of the NAD(P)-dependent epimerase/dehydratase family that is responsible for the last step of the major metabolic pathway resulting in GDP-L-fucose synthesis from GDP-D-mannose. It catalyzes a combined epimerase and NADPH-dependent reductase reaction, converting GDP-4-keto-6-D-deoxymannose to GDP-L-fucose. This reaction involves epimerization at both C-3 and C-5 followed by an NADPH-dependent reduction of the carbonyl at C-4. GDP-L-fucose synthase is also reported to act as the main autoantigen for a brain-infiltrating T cell clone. It is recognized by cerebrospinal fluid-infiltrating CD4+ T cells from HLA-DRB3*-positive subjects. Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected TSTA3 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected TSTA3 in MCF-7 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100-1,000 dilution from a representative lot detected TSTA3 in human testis and human kidney tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TSTA3 in HepG2 cells.

Affinity Binding Assay: A representative lot of this antibody bound TSTA3 peptide with a KD of 1.7 x 10-8 in an affinity binding assay.

Anti-TSTA3, clone 2N10-H1 ZooMAb®, Cat. No. ZRB1198, is a recombinant Rabbit monoclonal antibody that specifically targets TSTA3 and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry, and Western Blotting.

Biochem/physiol Actions

Clone 2N10-H1 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects human GDP-L-fucose synthase. It targets an epitope within 20 amino acids from the C-terminal region.

Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
