General description
CASP8 and FADD-like apoptosis regulator (UniProt: O15519; also known as Caspase homolog (CASH), Caspase-eight-related protein (Casper), Caspase-like apoptosis regulatory protein (CLARP), Cellular FLICE-like inhibitory protein (c-FLIP), FADD-like antiapoptotic molecule 1 (FLAME-1), Inhibitor of FLICE (I-FLICE), MACH-related inducer of toxicity (MRIT), Usurpin, CASP8 and FADD-like apoptosis regulator subunit p43, CASP8 and FADD-like apoptosis regulator subunit p12) is encoded by the CFLAR (also known as CASH, CASP8AP1, CLARP, MRIT) gene (Gene ID: 8837) in human. The CASP8 and FADD-like apoptosis regulator (FLIP) is a protein involved in modulating apoptosis and necroptosis pathways, primarily through its interaction with caspase-8 and FADD at the death-inducing signaling complex (DISC). Structurally similar to caspase-8, FLIP lacks enzymatic activity but serves as a crucial regulator of cell survival and death. It is expressed in various tissues, including lymphoid tissues, and plays a role in the TNFR1 and FAS-mediated signaling pathways. FLIP has multiple isoforms, including c-FLIPL and c-FLIPS, which influence its regulatory effects. While c-FLIPL can inhibit apoptosis by forming an inactive heterodimer with procaspase-8, c-FLIPS blocks the recruitment of caspase-8 to DISC. Dysregulation of FLIP is implicated in cancer progression, autoimmune disorders, and inflammatory diseases. It is considered a therapeutic target for conditions such as nonalcoholic steatohepatitis and hepatocellular carcinoma, where its modulation can impact cell death and inflammatory responses. Western Blotting Analysis: A 1:1,000 dilution of this antibody detected FLIP in A-431 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected FLIP in C2C12 cell lysate.
Affinity Binding Assay: A representative lot of this antibody bound with FLIP peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected FLIP in human skeletal muscle tissue sections.
