General description
Histone-lysine N-methyltransferase EHMT2 (UniProt: Q96KQ7; also known as Euchromatic histone-lysine N-methyltransferase 2, HLA-B-associated transcript 8, Histone H3-K9 methyltransferase 3 (H3-K9-HMTase 3), Lysine N-methyltransferase 1C, Protein G9a, EC 2.1.1.367) is encoded by the EHMT2 (also known as BAT8, C6orf30, G9A, KMT1C, NG36) gene (Gene ID: 10919) in human.
Histone-lysine N-methyltransferase EHMT2 (G9a) is a protein lysine methyltransferase that catalyzes mono- and di-methylation of histone H3 at lysine 9 (H3K9me1 and H3K9me2). This modification is associated with transcriptional repression and epigenetic regulation of gene expression. EHMT2 contains a cysteine-rich region (CRR) with an embedded RING domain, as revealed by X-ray crystallography, which plays a role in its catalytic activity and protein interactions. In addition to histones, EHMT2 methylates non-histone proteins such as p53, influencing cellular processes like tumorigenesis and apoptosis. EHMT2 is essential for embryogenesis, as genetic deletion in mouse models results in embryonic lethality. Its dysregulation is implicated in various cancers, including hepatocellular carcinoma, neuroblastoma, and lung cancer. EHMT2 contributes to tumorigenesis by epigenetically silencing tumor suppressor genes through promoter methylation. In hepatocellular carcinoma, EHMT2 suppresses APC expression, promoting Wnt/β-catenin pathway activation. Evaluated by Immunohistochemistry in human colon tissue sections.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected G9a in human colon tissue sections.
Tested Applications
Affinity Binding Assay: A representative lot of this antibody bound with G9a peptide with at least one hundred thousand-fold (100,000X) higher affinity than with non-specific control peptide.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected G9a in MCF7 cells.
Note: Actual optimal working dilutions must be determined by end user as specimens and experimental conditions may vary with the end user.
